MOTS-c (Mitochondrial Open Reading Frame of the 12S rRNA-c) is a mitochondrial-derived peptide composed of 16 amino acids, encoded by the mitochondrial genome. It plays a crucial role in regulating metabolic homeostasis, cellular stress responses, and insulin sensitivity. Unlike nuclear-encoded hormones, MOTS-C acts both in the cytoplasm and nucleus, influencing gene expression related to metabolism.

Preclinical studies have shown that MOTS-C activates AMP-activated protein kinase (AMPK) and enhances glucose utilization, mitochondrial function, and lipid metabolism. In a key animal study published in Cell Metabolism (2015), MOTS-C administration improved insulin sensitivity, protected against diet-induced obesity, and extended lifespan in mice on a high-fat diet. Early human data are limited but promising. A 2020 pilot study in older adults, published in Aging Cell, found that MOTS-C levels naturally decline with age and that exogenous MOTS-C administration enhanced physical performance and metabolic resilience.
