PT-141, also known as Bremelanotide, is a synthetic peptide derived from melanotan II, developed primarily for treating sexual dysfunction. Unlike PDE5 inhibitors like sildenafil (Viagra), PT-141 works through the melanocortin receptor pathway (MC4R) in the central nervous system, stimulating sexual arousal via the hypothalamus rather than affecting blood flow directly.

Although initially investigated for male erectile dysfunction, development shifted to focus on female sexual health. PT-141 is FDA-approved under the brand name Vyleesi for premenopausal women with hypoactive sexual desire disorder (HSDD). In Phase 3 clinical trials published in Obstetrics & Gynecology (2019), Bremelanotide significantly improved sexual desire scores, and reduced distress related to low libido, with effects seen in about 25% of treated participants compared to 17% on placebo.
